Abstract

MicroRNAs (miRNAs) represent an important class of noncoding RNAs that play key roles in regulating the expression of genes in health and disease. Accumulating experimental, pathological, genetic, epidemiological, and clinical studies highlight a growing list of miRNAs that control lipoprotein homeostasis and high-density lipoprotein (HDL) metabolism. By "fine-tuning" the expression of genes or signaling networks, miRNAs actively modulate HDL biogenesis and function including efflux of cholesterol from the vessel wall, altering inflammatory responses, or facilitating reverse cholesterol transport. This chapter highlights emerging concepts in miRNA-mediated control of HDL homeostasis as well as some of the challenges and therapeutic opportunities in the pathogenesis of cardiovascular and metabolic diseases.
